The Expression of TLR2 and TLR3 in Sertoli Cells of Azoospermic Patients

Objective: Toll like receptors (TLRs) placed on sertoli cells have very essential roles in sperm protection. This survey conducted to investigate the expression of TLR2, 3 genes in sertoli cells of men with azoospermia. Materials and methods: In this experimental study, testis biopsies were taken from 10 azoospermic men. The following enzymatic dissociation, the samples moved to lectin coated petri dishes. After a few passages, all cells were cultivated and seroli cells sorted by flow cytometry and then gene and protein expression of TLR 2, 3 were explored with RT- PCR and western blot, respectively. To confirm sertoli cells purification, alkaline phosphatase activity, immunohistochemistry assays were employed. Results: Isolation, purification and cultivation of the human sertoli cells were performed successfully. Efficacy of purification of sertoli cells by FACS sorter was ~97%. The type of cultured cells was confirmed by vimentin and FSH receptor markers. Furthermore, the existence of anti-müllerian hormone in culture was confirmed. RT-PCR showed that TLR2, 3 genes were expressed in sertoli cells. Moreover, Q-PCR indicated that relative gene expression of TLR3 was significantly higher than TLR2 in sertoli cells. Meanwhile TLR2, 3 genes are expressed in fibroblast cells, but the level of expression is significantly less than in sertoli cells. Also TLR2, 3 proteins were expressed in sertoli cells. Conclusion: This study indicated expression of TLR2, 3 in human sertoli cells which could be indicator of their roles in developing immunity against pathogens as well as allo and auto antigens or viral antigens in seminiferous tubules.
