عنوان انگلیسی Expression of hsa-miR-204, RUNX2, PPARγ and BCL2 in bone marrow derived mesenchymal stem cells from multiple myeloma patients and normal individuals
چکیده انگلیسی مقاله Objective: Multiple Myeloma is a heterogeneous cytogenetically disorder in which clonal plasma cells proliferate in bone marrow and cause bone destruction. Bone marrow microenvironment play crucial role in pathogenesis of this disease and mesenchymal stem cells (MSCs) are one of the key players. Herein, we proposed to investigate the expression of hsa-miR-204, RUNX2, PPARγ and BCL2 as factors involved in osteogenesis, adipogenesis and MSCs survival in bone marrow derived mesenchymal stem cells from multiple myeloma patients and normal individuals. Materials and methods: MSCs were isolated from bone marrow aspirates of multiple myeloma patients and healthy donors and total RNA were extracted before and after co-culturing with L363 myeloma cells. The gene expression of RUNX2, PPARΓ, BCL2 and hsa-miR-204 were assessed using qRT-PCR method. Results: Higher level of RUNX2, PPARγ and hsa-miR-204 expression was observed in MM-MSCs compared to ND-MSCs. Although the expression of BCL2 showed decrease in MM-MSCs. Different results observed in co-culture model. Conclusion: Collectively, it can be concluded that MM-MSCs gene expression profile differ compared to ND-MSCs. MM-MSCs by up-regulation of RUNX2, PPARγ and hsa-miR-204 compared to ND-MSCs would result in bone formation defect. Also down-regulation of BCL2 would lead to MM-MSCs cell death.
