Liquid chromatography mass spectrometry-based metabolite pathway analyses of myeloma and lymphoma patients

Objective: To identify differentially expressed metabolites of Non-Hodgkin’s lymphoma and myeloma patients from healthy individuals. Materials and methods: Plasma samples were collected from eleven patients with the approval of the Ethics Committee of Health Service Center at Seoul National University: six are healthy controls (without blood cancers), and five patients with multiple myeloma (3 samples) and Non-Hodgkin’s Lymphoma (NHL) (2 samples) were available during the preliminary study period. Samples were analyzed using quadrupole time-of-flight liquid chromatography mass spectrometry and significant features generated after statistical analyses were used for metabolomics and pathway analysis. Results: Data after false discovery rate adjustment at q=0.05 of features showed 136 and 262 significant features in lymphoma patients while 350 and 98 features in myeloma patients for both positive and negative ionization modes respectively. KEGG pathway analysis has found pathways such as steroid hormone biosynthesis, ABC transporters, and arginine and proline metabolism to be affected in lymphoma patients. Myeloma patients, on the other hand, have pyrimidine metabolism, carbon metabolism, and bile secretion pathways to be potentially affected by the disease. Conclusion: The results show huge differences in the metabolites of healthy individuals from myeloma and lymphoma patients. Validation through quantitative metabolomics is encouraged especially for the metabolites that were seen to be significantly expressed in blood cancer patients.
