عنوان انگلیسی Enhanced DNA damages of human prostate cancer cells induced by radiofrequency capacitive hyperthermia pre and post X-rays: 6MV versus 15MV
چکیده انگلیسی مقاله Objective: The aim of this experimental study was to determine the effect of 13.56MHz radiofrequency (RF) capacitive hyperthermia, on radiosensivity of human prostate cancer cells pre and post X-radiation. Materials and methods: Human prostate cancer cells DU145, were cultured as spheroids in 300 micron diameter. The spheroids were treated in eight groups: group I: control, group II: RF hyperthermia at a temperature of 43°C for 30 minutes (HT), group III: irradiation at 4Gy by 6 MV photons (RT(6 MV)), group IV: irradiation at 4Gy by 15 MV photons (RT(15 MV)), group V: HT+RT(6 MV), group VI: HT+RT(15 MV), group VII: RT(6 MV)+HT and group VIII: RT(15 MV)+HT . DNA damages in term of tail moment were evaluated by alkaline comet assay. Thermal enhancement factor (TEF) was obtained for different treatment combinations. Results: Mean tail moment (TM) increased with increasing photon energy. In the HT group (II) TM was significantly larger than in group I. TM values in group III and IV were significantly higher than in group I. There were significant differences in TM between group V, VII and group III. Also, there were significant differences in TM between group VI, VIII, and group IV. TEF values demonstrated that enhanced response to radiation was more pronounced in group V compared with other combined treatments. Conclusion: Our results suggest that HT applied before RT leads to higher radiosensivity than reverse sequence. Hyperthermia at 43°C for 30 minutes added to 6 MV photons causes higher enhancement effect of radiation than 15 MV.
