عنوان انگلیسی The role of radiofrequency hyperthermia in the radiosensitization of human prostate cancer cell line
چکیده انگلیسی مقاله Objective: The aim of this study was the evaluation of enhanced induced DNA damages and apoptosis of prostate cancer cells, DU145, in spheroid culture, treated by combination of radiofrequency hyperthermia with 15 MV X-rays originated from external radiotherapy machine compared to radiation alone. Materials and methods: DU145 cells were cultured as spheroids in 300 micron diameters. Cultures were exposed to RF hyperthermia (HT) 90 minutes at 43 °C by RF waves were originated from a Celsius TCS system. Hyperthermia followed by external X-rays at doses of 2 Gy or 4 Gy in 15MV energy, with 15minutes intervals between them. The trypan blue exclusion assay, alkaline comet assay and Annexin V/PI flow cytometry, were performed to measure the viability, the amount of DNA damages in individual cell as tail moment and percentage of induced cell apoptosis, respectively in response to the combination of radiation and hyperthermia. Results: Viability, tail moment and the percentage of apoptosis values, were obtained after combination tratment of radiations and hyperthermia. Thermal enhancement factor (TEF), was calculated for combination treatment regime. Hyperthermia treatment followed by 4Gy of X-rays, resulted into the minimum viability (85.33± 1.3 %), highest tail moment (1.97± 0.1) and highest percentage of apoptotic cells (64.49±1.96%) rather than other treatments. The TEF’s were, 2.53and 2.33 from comet assay and flow-cytometry, respectively. Conclusion: The present data suggest that combination treatment of mega voltage X-ray and RF hyperthermia can result to a significant radiosensitization of prostatic cancer cells.
