عنوان انگلیسی Comparison of miRNA Profiles of Cord Blood Stem Cells in Identical and Fraternal Twins
چکیده انگلیسی مقاله Objective: Any difference in the intrauterine environment can affect the gene-expression profiles. MicroRNAs (miRNA) expression is regulated by epigenetic mechanisms. Differences in miRNA expression levels can be considered as a factor for discordance in identical twins. Regarding to the relationship between epigenetic and miRNA, twins is considered as a valuable model in determining the relative contribution of genetic and environmental factors. In this study, we compared discordance in expression level of 44 miRNAs in hematopoietic stem cells (HSCs) of identical and fraternal twins at birth. Materials and methods: Cord blood samples of identical and fraternal twins were collected in blood bags containing anti-coagulant and, CD133+ HSCs were isolated by magnetic-activated cell sorting (MACS) method. Differences in the gene expression levels of 44 miRNAs in CD133+ HSCs were evaluated using qRT-PCR. Results: In fraternal twins, 20 miRNAs had a high difference expression more than 10 fold and 10 miRNAs had a little difference in expression (less than 2-fold). Identical twins showed a high difference in expression of 13 miRNAs (more than 10-fold) and 18 miRNAs had a little discordance in expression (less than 2-fold). Conclusion: Identical twins had a positive correlation in miRNA expression, while the correlation was not statistically significant in fraternal twins. Altogether, more discordance in miRNA expression of fraternal twins can be attributed to both genetic and intrauterine environment; The Contribution of intrauterine environment and genetic in miRNA expression of HSCs was estimated 8% and 92%, respectively.
