Comparison of The Melatonin Preconditioning Efficacy between Bone Marrow and Adipose-Derived Mesenchymal Stem Cells

Objective: It is accepted that sources of mesenchymal stem cell might influence on the cell therapy and regenerative medicine outcomes. In other side, preconditioning strategy with different agents could improve the efficacy of cell therapy. Therefore, this study compares the cell protective effects of melatonin as as antiinflamatory and antioxidants agents on two types of mesenchymal stem cells; Bone marrow and Adipose tissue. Materials and methods: Rat BMMSCs and ADSCs were isolated and expanded. Preconditioning was performed with 5µM melatonin for 24 hours. Cell proliferation and viability were detected with MTT assay. Expression of Bax and Bcl2 genes, melatonin receptors and osteocalcin were performed with RT-PCR. Also, the apoptosis was detected with appropriate protocol. Osteogenic differentiation was analysed using alizarin red. Results: No significant increase was found in cell viability between BMMSCs and ADSCs after melatonin preconditioning. Melatonin down-regulated the Bax expression more in ADSCs compared to BMMSCs; however the decline was significant in both of them after preconditioning. Bcl2 expression was increased significantly in both two cell types; however, increase of Bcl2 expression was higher in ADSCs rather than in BMSCs. MT1 and MT2 was up regulated significantly in both cell types after preconditioning; but more in BMSCs. Melatonin increased osteogenesis capability with increase in osteocalcin expression. However, expression of osteocalcin in BMSCs before and after preconditioning was higher than that in ADSCs but melatonin increased it in ADSCs higher than that in BMSCs. Melatonin increased alizarin red concentration significantly in both BMMSCs and ADSCs (P< 0.05). Alizarin red staining severity increased significantly in ADSCs after preconditioning compared to BMMSCs (P< 0.05). Conclusion: It seems that preconditioning efficacy of melatonin in ADSCs is higher than that in BMMSCs. These findings could use in adoption of proper preconditioning protocol after considering the MSC source in specific clinical applications specially in bone regeneration.
