عنوان فارسی بررسی تاثیر پلی‌مورفیسم‌‌‌‌‌‌‌‌‌‌‌‌های تک نوکلئوتیدی ژن آدیپونکتین ۲۲۴۱۷۶۶rs، ۱۵۰۱۲۹۹ rs و ۱۷۳۰۰۵۳۹rs بر سن ابتلا به دیابت و سن شروع انسولین درمانی در بیماران مبتلا به دیابت نوع ۲
چکیده فارسی مقاله مقدمه:  ابتلا به دیابت نوع دو در سنین پایین در حال رشد است، احتمالا عوامل ژنتیکی نقش مهمی در شروع زودرس دیابت نوع دو دارند. هدف از مطالعه حاضر بررسی تاثیر پلی‌مورفیسم‌های تک نوکلئوتیدی ژن آدیپونکتین (T/G 45+)2241766rs، (G/T 276+)1501299 rs و (G/A 11391-) 17300539rs بر سن ابتلا به دیابت و سن شروع انسولین درمانی در بیماران مبتلا به دیابت نوع 2 بود. روش­ها: در این مطالعه موردی- شاهدی، 183 بیمار مبتلا به دیابت نوع دو و 155  فرد سالم شرکت کردند. داده‌های جمعیت شناختی و تن سنجی جمع‌آوری شد. بررسی ژنوتایپی پلی‌مورفیسم‌ها با استفاده از روش تقویت سیستم مقاوم به جهش واکنش زنجیره‌ای پلی‌مراز (T-ARMS-PCR) انجام شد. داده‌ها با استفاده از نرم افزارهای SPSS (نسخه 26) تجزیه و تحلیل شدند. یافته­ها: طبق نتایج، در  T/G45+ SNP سن ابتلا به دیابت و سن شروع انسولین درمانی در حاملین ژنوتایپ TG به طور معنی‌داری کمتر از حاملین ژنوتایپ GG بود. (به ترتیب 035/0= p، 048/0= p). در T/G45+ SNP ، 6/48 درصد از بیماران با سن ابتلای < 35 سال ، حامل ژنوتایپ TG بودند(013/0 p=). و ژنوتایپ TG (006/0=p، 994/7 –430/1=CI، 381/2=OR) به طور معنی‌داری با افزایش خطر ابتلا به دیابت در سن کمتر از 35 سال مرتبط بود. نتیجه­گیری: طبق نتایج پژوهش حاضر T/G45+ SNP از ژن آدیپونکتین اثر قابل توجهی در شروع زودهنگام T2DM و همچنین پیشرفت سریع بیماری دارد. بررسی واریانت های ژنتیکی موثر می‌تواند در غربالگری پیش از ابتلا  به منظور تشخیص زودهنگام و ارائه راهکارهای پیشگیرانه مفید باشند.

عنوان انگلیسی Investigation the effect of single nucleotide polymorphisms of adiponectin gene rs2241766, rs1501299 and rs17300539 on the age of diabetes onset and starting insulin therapy in patients with type 2 diabetes
چکیده انگلیسی مقاله Research aim: The incidence of type 2 diabetes is growing at an early age. Genetic factors probably play an important role in the early onset of type 2 diabetes. The aim of the present study was investigation the effect of single nucleotide polymorphisms of adiponectin gene rs2241766 (+45 T/G), rs1501299 (+276 G/T) and rs17300539 (-11391G/A) on the age of diabetes onset and starting insulin therapy in patients with type 2 diabetes. Methods: In this case-control study, 183 patients with type 2 diabetes and 155 healthy individuals participated. Demographic and anthropometric data were collected. Genotyping was performed by using amplification refractory mutation system-PCR (ARMS-PCR). Data were analyzed by using SPSS software (version 26). Results: According to results, in rs2241766 (SNP+45 T/G), the age of onset of diabetes and age of starting insulin therapy in the TG genotype carriers was significantly less than in TT genotype carriers (Respectively P = 0.035, P=0.048). In SNP+45 T/G, 48.6% of patients with diabetes onset age < 35 years were carrying the TG genotype (p = 0.013). TG genotype (OR=2.381, 95% CI:1.430–7.994, p = 0.006) were associated with the significantly increased risk of diabetes onset age < 35 years. Conclusion: According to the results of the present study, SNP+45 T/G of the adiponectin gene has a significant effect on the early onset of T2DM as well as the rapid progression of the disease. Evaluation of effective genetic variants can be useful in pre-infection screening for early detection and prevention.
