عنوان انگلیسی Apoptotic effect of Phycocyanin on HT-29 colon cancer through activation of caspase family and P53 cell signaling pathway
چکیده انگلیسی مقاله Background: The C-phycocyanin(C-PC) from Spirulina platensis is a future candidate of cancer treatment; C-PC originated from edible blue-green algae and as a biliprotein, numerous in vivo and in vitro research has reported its anti-cancer activity. In summary, the effect of C-PC on caspase 3, 8, 9, and p53 pathways in  the human colorectal adenocarcinoma cell line (HT-29) and human umbilical vein endothelial cells (HUVECs) is investigated.
Materials and methods: In the current research, we investigate the effect of C-phycocyanin on caspase 3, 8, 9, and p53-mediated apoptosis pathways in two cell lines (HT-29 and HUVEC) using quantitative real-time PCR (qRT-PCR) and Flow cytometry assay. Moreover, the cytotoxicity of phycocyanin on HT-29 cells was compared with HUVEC normal cells via colorimetric assay.
Results: Based on our findings, expression of caspases 3, 8, 9, and p53 genes were increased in the colorectal cancer cell treated with C-PC (P<0.01**); these molecular results were confirmed by an increase in the number of Colorectal cancer cells in the early and late stages of apoptosis as compared to untreated cells. In addition, the results of the MTT colorimetric assay showed that C-PC has not cytotoxicity effects on HUVECs cells (normal cell) (P<0.01**).
Conclusion: Collecting data appears that C-PC has an appropriate effect on apoptosis. Since tumorigenesis is halted by apoptosis, C-PC can be a hopeful product in colorectal cancer treatment.
