Toxicity of Antibiofilm Alginate Lyase from Streptomyces olivaceus on Wistar Rats (Rattus norvegicus)

Background:
The alginate lyase is known to have antibiofilm activity. Toxicity tests for the alginate lyase produced by
Streptomyces olivaceus
originating from Serena Kecil Island, North Sulawesi, Indonesia have never been reported. This research conducted an in vivo toxicity study of the alginate lyase using Wistar rats for its development as an oral anti-biofilm agent.
Methods:
Twenty male Wistar rats were divided equally into a technical control (TC) group which was only given food and drink, a negative control (NC) which was given Phosphate Buffer Saline 1x, an experimental group (E1) which was given alginate lyase at a dose of 0.63 g/kgBW, and (E2) which was given alginate lyase at a dose of 20.85 g/kgBW. Toxicity tests were carried out for 7 days by observing the parameters of changes in body weight and behavior of rats, changes in AST and ALT enzyme levels, and evaluation of macroscopic and microscopic damage of the liver.
Results:
After 7 days of treatment, rats in the NC, E1, and E2 groups experienced insignificant weight loss, also had normal behavior and did not experience symptoms of toxicity or death. AST and ALT levels in rats which was given alginate lyase (E1 and E2) decreased significantly but were still within the normal range. Alginate lyase also causes an adaptive stress response and parenchyma degeneration of liver cells.
Conclusion:
Alginate lyase at a dose of 0.63 g/kgBW and a dose of 20.85 g/kgBW were considered safe for rats (
Rattus norvegicus
) and has the prospect of being developed as an anti-biofilm agent.
