عنوان انگلیسی Identification and validation of tumorigenesis related hub genes in hepatocellular carcinoma via integrated bioinformatics analysis
چکیده انگلیسی مقاله Hepatocellular carcinoma (HCC) is a heterogeneous and complex liver cancer with poor prognosis and high recurrence rates. Current research lacks consistency, and the molecular mechanisms driving tumorigenesis across various etiologies and geographic regions are not well understood. To address this, we integrated large cohort genomic data to identify distinct gene expression patterns linked to different causes of HCC. We performed differential expression analysis on genomic datasets from patients with HCC and precancerous conditions, revealing key differentially expressed genes (DEGs) such as CBS, ELF3, SPP2, and RPL8. These DEGs were correlated with overall survival in HCC patients. Enrichment analysis based on the Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways identified significant life activity pathways involved in HCC development. We found more DEGs between liver cirrhosis and tumor than between hepatitis and tumor, suggesting cirrhosis plays a more prominent role in HCC progression. A 47-gene set was identified as a potential ‘brake’ for benign-to-malignant liver transformation. Notably, the PI3K-Akt signaling pathway was the top enriched pathway, indicating its central role in the transition from normal liver to cirrhosis and then to tumor. These findings highlight potential prognostic and therapeutic biomarkers for different HCC subgroups.
