Is-1: Antigenicity and Immunogenicity of Prostate Cancer Stem-Like Cells

Objective: While consistent evidence supports the concept that, irrespective of the cell-of-origin, many cancers are hierarchically organized as a normal tissue, and therefore, they contain stem-like/progenitor cells (CSCs), whether CSCs are susceptible to immune surveillance is poorly defined. We addressed this biological problem in a mouse model of prostate cancer. Materials and Methods: Utilizing the sphere assay, we established long-term prostate CSC lines from unsorted prostate cells obtained from different stages of prostate cancer progression in transgenic adenocarcinoma of the mouse (TRAMP) mice, which develop autochthonous prostate lesions evolving from mouse prostate intraepithelial neoplasia (mPIN) to NE differentiation. CSCs lines were assessed in vitro and in vivo for their antigenic content and immunogenicity, i.e., the ability to induce a specific immune response. Results: Prostate CSC lines expressed prostate cancer associated antigens, MHC I and MHC II molecules and ligands for natural killer (NK) cells. Indeed, CSCs were targets of NK and cytotoxic T lymphocytes both in vitro and in vivo. Vaccination with dendritic cells pulsed with irradiated CSCs induced a tumor-specific immune response that was stronger than the one induced by dendritic cells pulsed with differentiated tumor cells, delayed tumor growth in mice challenged with prostate CSCs, and caused tumor regression in TRAMP mice. Conclusion: Thus, CSCs are targets of both innate and adaptive immune responses and might be exploited for the design of novel immunotherapeutic approaches against cancer.
