عنوان انگلیسی Ps-44: Effect of Rosiglitazon on Expression of Fibronectin Type III Domain- Containing 5 Protein and Cardiac Differentiation
چکیده انگلیسی مقاله Objective: Fibronectin type III domain-containing 5 protein (Fndc5) is a type I membrane protein that has 209 amino acid residues. Bostrom et al. found that Peroxisome proliferator-activated receptor (PPAR) γ coactivator 1α (PGC1α) transcriptional co-activator expression induced Fndc5 expression in white adipose tissue (WAT) browning. Previous studies by our group have shown that Fndc5 expression increased during the process of cardiac differentiation of mouse embryonic stem cells (mESCs), leading us to conclude that it might be involved in cardiogenesis. Also we showed decreased PGC1α expression significantly reduced expression of Fndc5 and cardiac markers in cardiomyocyte. Materials and Methods: In the present study, correlation between PGC1α and Fndc5 was examined by application of a PGC1α agonist (Rosiglitason) that increased PGC1α expression. Results: Our results indicated that in the process of beating cardiomyocyte formation, Fndc5 and cardiac markers expression level was not influenced by PGC1α agonist significantly. Conclusion: These results propose an idea that the ground state of PGC1α expression level is needed which needs further verification.
