عنوان انگلیسی Ps-155: Transdifferentiation of Bone Marrow Stromal Cells into Cholinergic- Like Cells by Nerve Growth Factor and β-Mercaptoethanol
چکیده انگلیسی مقاله Objective: Bone marrow mesenchymal stem cells (BM-MSCs) are pluripotent cells that have the ability of division, self-renewal and creation of a variety of differentiated cells such as neurons that can be used as a source of cells to repair damaged tissues. The aim of this study was to investigate the effects of nerve growth factor (NGF) and bone marrow stromal cells (BMSCs) on differentiation of BM-MSCs into cholinergic neurons by evaluation of OCT4 and Neuro D expression. Materials and Methods: Mesenchymal stromal cells were isolated from bone marrow of adult rat long bone and cultured in α -MEM medium containing 10% FBS. After 5 passages, the expression of OCT4 and Neuro D was evaluated using PCR. The cells were preinduced by BME for 1 day followed by induction with NGF for 29 days. Data was analyzed using ANOVA and Tukey post test. Results: Expression of pluripotency marker Oct4 was increased during the pre- induction phase and decreased during the induction period,whereas Neuro D expression was not seen in pre-induction phase, but its expression increased along time in induction period. Conclusion: These findings showed reasonable harmony between the increased expression of NeuroD and decreased expression of Oct4, that approved selected way to the differentiation of BM-MSCs to cholinergiclike cells.
