عنوان انگلیسی Ps-10: The Effect of Bisphenol A on Osteogenic Activity and Morphology of Rat Bone Marrow Mesenchymal Stem Cells In Vitro
چکیده انگلیسی مقاله Objective: Bisphenol A known to be a xenoestrogen, is widely used in industry and dentistry. In addition to environmental problems, also can be harmful to human health.This study was aimed to evaluate the effect of bisphenol A (BPA) on the viability and osteogenic differentiation in rat bone marrow mesenchymal stem cells to osteoblast. Materials and Methods: Rat bone marrow mesenchymal stem cells were extracted under strile conditions using flashing-out method. At the end of third passage, cells were divided to 2 groups control and bisphenol A (250nM), for a period of 21 days in the osteogenic media (DMEM containing 10% fetal bovine serum, 10 mM β-glycerol phosphate, 10 nM dexamethasone and 50 g/ ml ascorbic 3-phosphate). The cell proliferation and viability, bone matrix mineralization, intercellular calcium deposition, extracellular calcium deposition, alkalin phosphatase activity and morphologic changes of the cells were measured using MTT assay, alizarin red staining, calcium kit, von kossa staining, alkalin phosphatase kit and Fluorescence staining respectively. Data was analyzed using way ANOVA and means difference was considered significanat at p< 0.05. Results: A significant reduction in the cell viability, mineralization and calcium deposition , alkalin phosphatase activity,change of chromatin condensation, and cytoplasm morphology was seen in the cells treated with bisohenol A. whereas These parameters were significantly increased in the control level.Conclusion: Since Bisphenol A caused a significant decrease in viability, osteogenic differentiation in the rat bone marrow mesenchymal stem cells, therefore It can be a factor in the development of osteoporosis.
