P-4: Oxidant/Antioxidant Imbalance in Converting Fibroblast Dominant Condition to Epithelial Tissue Formation in Long-TermHuman Skin Culture

Objective: In attempt to isolate multi-potent stem cells from foreskin we encountered an interesting phenomenon, converting fibroblast dominant condition to epithelial tissue overgrowth in long-term culture. However, the basic mechanism(s) which may be involved in this conversion is not clear. This study was designed to examine the role of oxidant/antioxidant system in this phenomenon. Materials and Methods: The explanted tissues were obtained by spreading out the small sized foreskin derived tissue onto the cell culture dishes. After a 40 - day period, glucose, total protein, antioxidant capacity and protein profiles were determined in the cell culture supernatant and compared to DMEM supplemented with glutamine, antibiotics and FBS as base line medium. Also the dishes were stained by routine H&E. Results: Fibroblast and uncharacterized fibers emerged from beneath of the specimen firstly and gradually overgrew within the first month. Surprisingly these cells began to disappear around day 30 while epithelial cells turned out to be the major cells .Ultimately on day 40 the epithelial tissue appeared. Total protein concentration were 1.44 mg/dl ,0.97 mg/dl ,concentrations of Glucose were 1.6 and 119.2 mg/dl for old and baseline medium respectively. The anti-oxidant activity of old medium was176.29 μM, in comparison to baseline medium 96.63 μM. The density of some proteins was higher for old medium corresponding to the 40-day culture. However there was no difference in protein pattern between the two media on SDS-PAGE. Conclusion: As our knowledge we are the first group showing that epithelial cell could overgrowth instead of fibroblast in long term foreskin tissue culture. This phenomenon may be due to cell injury induced by oxidant/antioxidant imbalances.
