چکیده انگلیسی مقاله Objective: Cryopreservation of mesenchymal stem cells (MSCs) from equine umbilical cord blood (EUCB) has been an important section of the cell culture, tissue engineering, and cell-based therapy. MSCs from umbilical cord blood (UCB) offer many advantages when used as grafts in cell transplantation because of the immaturity of new born cells. The use of MSCs in cell therapy has been developed in equine as well as in humans; therefore, preservation of these cells is of utmost importance in the future studies related to repair therapies. This preservation calls for defining requirements under which these cells may be able to tolerate the cryopreservation situations. The objective of the present study is to evaluate the viability ratio of the frozen/ thawed MSCs. Materials and Method: Materials and Methods cells were isolated from equine UCB soon after parturition and MSCs were purified after different subcultures. Purified cells from the 1st to 10th passages were frozen and stored in liquid nitrogen. Cell viability was assessed after thawing through trypan blue staining procedure. Results: High viability (>80%) ratio was observed with the cells derived from the 1st passage after one week and eight weeks of cryopreservation, thus, showing no effect of time. Conclusion: The results of the present study demonstrated that equine umbilical cord blood stem cells could successfully be frozen and stored in liquid nitrogen without much change in the characterization of the cells as regards their viability, growth ability and differentiation potential. However, the viability of cryopreserved cells may vary depending on the passages used for cryopreservation.
