چکیده انگلیسی مقاله Objective: Photodynamic therapy (PDT) is a therapeutic modality in cancer which uses visible light to activate a photosensitizer producing singlet oxygen in target cells or tissues. Recently, the attention on photosensitizers has been focused on porphyrins and their analogs and there have been many efforts into the synthesis of tetra-pyrrole macrocycles such as phthalocyanines and porphyrazines. The absorption in the high wavelength, high quantum yield, high triplet time and effective oxygen produce capacity and water solubility of these photosensitizers lead these complexes to have potential applications in photodynamic therapy of cancer. In the present study, the cytotoxic effect of N, N', N", N'''- tetramethyltetra-3, 4-pyridinoporphyrazinatozinc (II) [Zn (3,4-tmtppa)]4+ was assessed on adenocarcinoma cell line (HeLa) to evaluate its photodynamic efficiency. Materials and Methods: In order to measure cytotoxic effect of Zn (II) porphyrazine complex, a MTT assay was performed under three different conditions: no light, with light and with light and filter. The cells were treated with 0, 1, 6, 12 and 50 μM concentrations of the porphyrazine complex. Irradiation occurred with 150-W halogen lamp and with the aid of filter, the wavelength range was kept between 600- 800 nm. Results: Photo cytotoxicy of the porphyrazine was remarkably more than dark cytotoxycity. In addition, photo cytotoxicity increased when the filter was used. In the range of 0-12 μM, an increase in concentration of the complex resulted in increased cytotoxic effect, while at the higher concentration (50μM) cytotoxicity decreased probably due to the aggregation phenomenon. Conclusion: The porphyrazines [Zn (II) porphyrazine] are promising water soluble photosensitizers for innovative photodynamic therapy and present a high potential of application in treatment of cancer
