چکیده انگلیسی مقاله Objective: Zoledronic acid is a biphosphonate that is used to prevent skeletal fractures in patients with cancers, as well as for treating osteoporosis. It inhibit the osteoclast proliferation whereas stimulate the formation of osteoblast. Altogether, the mechanism of zoledronic acid in osteoblastic differentiation and gene expression is not fully understood. In this research, we evaluate effect of zoledronic acid on osteoblastic differentiation acceleration and bon sialoprotein expression. Materials and Methods: MSCs were isolated from human bone marrow. For osteogenic differentiation, hMSC were pulse treated with 5μM ZA for 3h, and incubated after a medium change in osteogenic differentiation medium for 3 weeks. RNA was extracted after first, second and third weeks of culture and also from undifferentiated MSCs. After cDNA synthesis, BSP expression was evaluated by quantitative real-Time PCR. Results: BSP expression sharply increased during osteoblastic differentiation of MSCs treated with zoledronic acid. BSP expression during osteogenic differentiation, compared to the undifferentiated MSCs, exhibited 305-fold, 1113-fold and 2023-fold increase in expression in 1, 2, 3 weeks of differentiation, respectively. Alizarin Red Staining confirmed the increasment of calcium deposits, characteristic of osteogenic cells in osteoblastic differentiated MSCs treated with zoledronic acid. Conclusion: Previous finding in osteoblastic differentiation of MSCs without zoledronic acid present that BSP expression during differentiation is low. Whereas, BSP expression during differentiation of MSCs treated with zoledronic acid is high. Therefore, Pulse treatment with zoledronic acid accelerate osteoblastic differentiation of mesenchymal stem cells and the increase of BSP expression confirm it.
