P-32: Histopathological Study of Effect of Essential Oil and Aqueous Extracts of Rosemary(Rosmarinus Officinalis) on Wound Healing of Diabetic BALB/c Mice

Objective: Rosemary (Rosmarinus officinalis) in traditional medicine is used to treat rheumatism, migraines, skin disease such as hair loss, infections and wound healing. Materials and Methods: Twenty male BALB /C mice were selected. In fifteen mice alloxan monohydrate was used for induction of diabetes. Five mice served as controls. In all, three full-thickness round wounds (4mm diameter) were created experimentally. then were divided into four experimental groups: 1 - control group (healthy - no treatment) 2 - diabetic - without treatment 3 - diabetic treated with 0.5 ml aqueous rosemary for 3 days via intraperitoneal 4 - diabetic and treated topically with essential oil made from rosemary plants amount to 20 microliter per each wound twice a day for three days. Wounds were evaluated macroscopically for 15 days. The blood glucose levels, body weight and rate of wound contraction at day 5 and 10 and 15 evaluated. 5th and 15th day after the injury, two mice in each group randomly selected and after necropsy histopathological studies were placed. Results: In groups 2 and 3 and 4 increased glucose level compared to group 1 was observed during the experimental period. But in groups 3 and 4, significant decrease in blood-glucose level on days 10 and 15 after wounding compared with group 2was seen. This reduction in 4 compared to 3 per day five were higher. Groups 2 and 3 and 4 suffered weight loss during the period, but in group 3 and 4 weight were reached significantly on days 10 and 15 in normal. This increase in group 4 than group 3 was more significant. In groups 3 and 4 histopathological study was detected reduced inflammation, re-epithelialization, and further contraction of wounds, angiogenesis and collagen deposition. Conclusion: Antioxidant effects of rosemary plants protect beta cells from more damage that is caused by alloxan and can facilitate the regeneration operation, which lead to increasing insulin secretion. This combination accelerates wound healing in diabetic mice. Through which the impact on various aspects of the healing process such as reduce inflammation, debridement, wound contraction and epidermal regeneration.
