P-33: Immunohistochemichal Study of Estrogen Receptor (ER), C-erbB-2 and Topoisomerase IIβ Binding Protein 1 (TopBP1) in Normal and Neoplastic Feline Mammary Tissue

Objective: In cats, the involvement of steroid hormones in the development of mammary carcinomas is supported by the protective effects of ovariectomy. C-erbB-2/neu is a transmembrane glycosylated protein that belongs to the human epidermal growth factor receptor (tyrosine kinase receptor) family and plays an important role in cell cycle regulation and differentiation. Topoisomerase IIβ binding protein 1 (TopBP1) has structural and functional similarities to the familial breast cancer gene BRCA1. Materials and Methods: Feline mammary tumors (n = 27) and normal tissues (n=5) were obtained.carsinamas were classified according to World Health Organization (WHO) criteria. Malignant tumours were graded as low (1), intermediate (2) or high (3) grade. For each sample, sections (5 μm) were stained with haematoxylin and eosin (HE) and Immunohistochemical labelling was performed using the avidin-biotin-peroxidase complex (ABC) procedure. Statistical analysis was performed by SPSS. Results: In compare with normal and neoplastis tissues, ER expression was significantly higher in normal tissues, hyperplasic/dysplastic lesions, and benign tumours respectively than in carcinomas. C-erbB-2 over expression was detected in only 1 of the 11 carcinoma (9. %) and did not correlate with histological classification. Two of the 6 (6.8%) hyperplastic or dysplastic lesions and one of the 10 (10%) mammary adenomas were C-erbB-2 positive. There was positive staining for TopBP1 in 26of 27 feline mammary lesions, the percentage of cells positive for TopBP1 increased with histological grade. Most staining was nuclear but both nuclear and cytoplasmic staining was observed as the degree of malignancy increased. Many neoplasms which are ER negative show TopBP1 immunoreactivity. Conclusion: The low percentage of ER-positive invasive carcinomas further demonstrated the aggressive phenotype and behavior of feline mammary tumours. C-erbB-2 may not play as significant role in mammary carcinogenesis and prognosis in cats as it does in human patients. The BRCA1-related protein TopBP1 is expressed in feline mammary tissues, with a higher percentage of cells staining in more malignant neoplasms compared to lower grade neoplasms and non-neoplastic lesions. The staining pattern shows a slight shift from nuclear to both nuclear and cytoplasmic staining in more malignant neoplasms.
