P-41: Anti Cancer Effect of Cyclodextrin Nanoparticles Containing Curcumin on hTERT Gene Expression in Breast Cancer Cell Line

Objective: Telomerase enzyme is expressed in most human cancers, including breast cancers. Breast cancer is the second common cancer worldwide. Targeting telomerase is effective way to overcome breast cancer. Curcumin, a hydrophobic polyphenolic compound derived from the rhizome of the herb Curcuma longa, possesses a wide range of biological applications including cancer therapy and special targeting telomerase. However, its prominent application in cancer treatment is limited due to sub-optimal pharmacokinetics and poor bioavailability. In order to improve its hydrophilic and drug delivery, we have utilized a β-cyclodextrin (CD) mediated curcumin drug delivery system via encapsulation technique. Materials and Methods: Curcumin encapsulated into cyclodextrin and the rate of curcumin loading into cyclodextrin was estimated. MTT assay was utilized to evaluate cytotoxicity effects of cyclodextrin-curcumin and free curcumin, then the cells were treated with equal concentration of cyclodextrin-curcumin and free curcumin. The last by Real-time PCR the rate of telomerase gene expression in groups were compared. Results: MTT assay demonstrated that β-cyclodextrincurcumin enhanced curcumin delivery in T47D breast cancer cells about 1.3 fold higher than free curcumin. Data analysis with SPSS showed that the rate of telomerase gene expression in equivalent concentrations with cyclodextrin-curcumin was lower than free curcumin in comparison (p=0.001). Conclusion: Results are suggesting that cyclodextrincurcumin complex can be more effective than free curcumin on the telomerase, because expression of telomerase gene was more reduced. Briefly, cyclodextrin-curcumin complex can use for target therapy of breast cancer.
