عنوان انگلیسی P-49: The Effect of CD11c+ CD8α+ Dendritic Cells Loaded with Tumor Lysate on Peritoneal Macrophages in Fibrosarcoma Bearing Mice
چکیده انگلیسی مقاله Objective: The use of dendritic cells (DCs) as a cellular adjuvant provides a promising approach in cancer immunotherapy. We investigated the effect of the CD11c+ CD8α+ DCs loaded with Tumor Lysate (TL) in cancer immunotherapy. Materials and Methods: The Balb/c mice were injected subcutaneously in the right flank with mouse Fibrosarcoma cells (Wehi-164) ten days before immunization with the CD11c+ CD8α+ DCs. The mice CD11c+ CD8α+ DCs were then isolated by magnetic cell sorting (MACS), then the TL pulsed CD11c+ CD8α+ DCs, un-pulsed CD11c+ CD8α+ DCs, TL and PBS as control group were injected, respectively. Tumor growth rate, survival and nitric oxide (NO) production were assessed. Results: Immunotherapy with TL- CD11c+ CD8α+ DCs and CD11c+ CD8α+ DCs led to a significant increase in the peritoneal macrophages NO production. Moreover, TL- CD11c+ CD8α+ DCs significantly induced the tumor growth suppression and these mice longer survival than the other immunized mice. Conclusion: The current study suggests an anti-tumor innate immunity against the Fibrosarcoma could be induced by TL- CD11c+ CD8α+ DCs and might provide useful therapeutic approach for cancer management.
