چکیده انگلیسی مقاله Objective: Recently, dendrimers have been widely used in medical applications such as drug delivery and gene transfection. Specially some therapeutic agents with hydrophobic structure could bypass their low waterinsolubility and bioavailability by encapsulating with them. In this study we attempt to determine inhibitory effect of polyamidoamine (PAMAM) dendrimer encapsulating curcumin on cancer cell proliferation through downregulation of telomerase. Materials and Methods: The cytotoxicity of PAMAM encapsulating curcumin was investigated by 3-(4,5- dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. Results: Dendrimer had no cytotoxicity by its own, also it showed dose-dependency with not any significant time-dependency for PAMAM encapsulating curcumin on T47D cell line of human breast cancer. Since for 24 hours, IC50= 10.5μM, we used 1, 2, 5 and 10μM for 24 hours treatment to aim therapeutic effect. To investigate anti-proliferation effect we used Telomerase Repeat Amplification Protocol (TRAP) assay and determined relative telomerase activity (RTA %) for each sample. Obtained RTA % decrease while PAMAM encapsulating Curcumin concentration increase. It suggests that PAMAM encapsulating curcumin have a dose-dependent cytotoxicity effect on T47D cell line through downregulationg and inactivation of telomerase and inducing apoptosis by enhancing curcumin uptake by cells. Conclusion: PAMAM could be considered as a good carrier specially for these kind of hydrophobic agents.
