P-58: Genetically Modified Stem Cells, a New Successful Way to Colon Cancer Treatment

Objective: "Wnts" are a large family of secreted glycoprotein ligands that regulate diverse processes through their interaction with their receptors on target cells. Aberrant activation of Wnt signaling pathway is the main driving force related to colon cancers, so that ~80% of colon cancers are associated with defects in this pathway. The colon cancer cells with high Wnt activity exhibit stem cell properties so that they have ability to repopulate a tumor after treatment. As high activation of Wnt signaling pathway is one of the major mechanisms of tumorigenesis in colon, identification of inhibitors of the pathway may be useful in colon cancer therapy. Materials and Methods: Investigations aimed to discover new Wnt antagonists have recently been started at Genetics Group of Biological Sciences Faculty, Tarbiat Modares University and the ability of this Wnt antagonists to inhibit Wnt signaling pathway activity was determined by an assay based on the expression of green fluorescent protein ORF inserted just in the downstream of the promoter of β-catenin gene normally regulated by Wnt signaling pathway. Results: The activation of Wnt signaling pathway was found to be negatively regulated by Wnt antagonists secreted from specific stem cells. Conclusion: Injection of specialized lines of stem cells secreting Wnt antagonists into tumor mass during surgery will improve the treatment of colon cancer. To this end, stem cells were genetically modified to specialized stem cells which are grafted in vivo and continue to express and secret soluble Wnt antagonists into the tumor mass containing cancer stem cells.
