عنوان انگلیسی P-60: Effect of Decapeptyl on Neurons in Hippocampus Area of Mice Affected by Chemotherapy with Cyclophosphamide
چکیده انگلیسی مقاله Objective: Gonadotropine hormone (GnRH) is the first reproductive key hormone. Today, GnRH analogues are widely used in vitro fertilization and treatment of sex hormone-depended cancers induced by the materials used in chemotherapy. Therefore, the aim of the present study is to evaluate the effect of cyclophosphamide and decapeptyl (analogues of GnRH) on neurons of hippocampus area in male mice. Materials and Methods: In this study, 24 male mice with same weights, after compromising with the experimental conditions, were divided in four groups as follows: the first treatment, cyclophosphamide (dose 65 mg/k), second treatment, decapeptyl (1.4 μg/k), third treatment, decapeptyl at first, and after 10 days cyclophosphamide was injected intraperitoneally. The control group was injected only with sterile saline. In order to evaluate the tissue changes in hippocampus area of mice, tissue sections were prepared after a month and stained with eosine, hematoxylin and PAS staining. Also, serum samples were obtained in order to measure the amounts of hormones FSH, LH and testosterone using ELISA kit. Results: The results showed that intraperitoneal injection of cyclophosphamide and decapeptyl in mice hippocampus results in tissue changes. In mice that had received cyclophosphamide, nuclei of neurons were being densed, their cytoplasm were changed and LH levels were decreased. But in the mice that had received decapeptyl, cell divisions were significant in neurons. LH and FSH levels showed a reduction and Testosterone showed an increase. Finally, the third group that had received both substances, the mentioned changes in the hippocampus region were observed. Conclusion: Based on these results, we can conclude that decapeptyl has a positive effect on hippocampus area neurons
