Objective: Human adipose tissue may ideal large-scale source for clinical application of neurodegenerative diseases. The aim of this study was to characterization and differentiation of human adipose-derived stem cells (ADSCs) into Schwann- like cells Materials and Methods: Human adipose tissue was obtained from 5 samples. ADSCs were isolated and then characterized by flowcytometry techniques for CD34, CD44, CD45, and CD90 markers. Adipose -derived stem cells grew in suspension as spheres in the presence of the mitogens factors. They were induced to differentiate and expression of the Schwann specific markers, GFAP and S100 was determined by Real time- PCR and Immunostaining techniques. In addition, MTT assay was used for detecting Percentage of cell viability. Results: Flow cytometry analysis of isolated cells showed that mesenchymal stem cell specific markers (CD90 and CD44) were positive but for other markers (CD45 and CD34) were negative. Using neurospheres forming protocol, the spheres have been able to proliferate and to differentiate into Schwannlike cells. After induction, the ADSCs changed from flat, elongated, to Schwann cell shape. Both Real time -PCR and Immunostaing analysis demonstrated differentiated cells strongly, expression of Schwannspecific markers. Conclusion: These findings indicated that ADSCs have a high potential for differentiation into Schwann cell. Therefore, ADSCs a favourable choice for the treatment of neurological diseases.
