P-77: Evaluation of Cytotoxicity Induced by 2 Resin Based Sealers (“2seal and AH-Plus®”) in Osteoblast Cell Lines

Objective: The aim of this study was to evaluate the cytotoxicity induced by 2 resin based sealers “2Seal and AH-Plus®” in 2 osteoblast- like cell lines “MG-63 and Saos-2”. Materials and Methods: Using sterile disks of both sealers in complete media, 24 and 72 hours extracts were prepared. The extracts were transferred to Saos-2 or MG-63 cell culture plates (already confluenced by 75%) and after 24 hours incubation, the cell viability tests were performed for each extract and cell line using MTT assay and trypan blue dye exclusion. Respective complete media was used as control group. Data were analyzed using Tukey HSD and one way analysis of variance (ANOVA). P< 0.05 was considered as the level of significance. Results: For both 24 and 72 hours extracts, AHplus and 2Seal showed cytotoxicity in both 2 cell lines. In Saos-2 cell line both sealers 24 and 72 hours extracts, did not show statistically different toxicity (p>0.05). In MG-63 cell line, 2Seal 24 hours extract and AHplus 72 hours extract showed more cytotoxicity compared to the other one, respectively (p< 0.05). Conclusion: AH-Plus and 2Seal both showed significant cytotoxicity in these two cell lines. While AH-Plus 72 hours extract cytotoxicity in MG-63 cell line had increased compared to 24 hours one, 2Seal showed no different cytotoxicity by its 72 hours and 24 hours extract. AH-Plus and 2Seal 24 hours extracts showed different level of cytotoxicity in MG-63 cell line while in Saos-2 cell line there was no detectable difference between these two sealers extracts. This might reflect the higher sensitivity of MG-63 cell line toward cytotoxicity induced by these two sealers compared to Saos-2.
