Cervical cancer is characterized by progression of premalignant lesions to malignant tumours. Approximately 500,000 new cases are diagnosed every year out of which 1/5 arise in India. Despite association of large number of risk factors, human papilloma virus (HPV) is found to be associated with more than 95% of the cases. Though important, studies suggest that HPV infection alone is not sufficient for tumorigenesis but many genetic factors are also required for the initiation and progression of the disease. WWOX a proapoptotic and a tumour suppressor gene, residing on second most common fragile site, interacts with p53 & p73 and induce apoptosis synergistically. Down regulation of WWOX protein was observed in various cancers e.g. breast, lung, ovarian, prostate, gastric, etc. Taking this into account, present study was designed to evaluate the expression of WWOX in cervical cancer. Analysis of the results showed a substantial down regulation of WWOX protein in invasive cervical cancer cases showing it may act as a tumour suppressor but results at transcriptional level were counterintuitive. The important aspect in such case is the aid provided by the host to the pathogen for manifesting itself. It came into light that the epitheliotropic nature of the virus is promoted by the host transcription factor, BRN3A, in the uterine cervix. Available literature provides the information that it activates the transcription of the genes of the high risk HPVs. In this study, the activity of BRN3A has been traced, where it interacts with different components of the cell facilitating the process of cell- proliferation. In order to understand the same, HPV-positive cervical cancer cells were subjected to chemotherapeutic (Cisplatin) treatment. Besides the activity of BRN3A, this experiment also revealed that BRN3A remains unaltered by the chemotherapy. Taking these into consideration along with its anti-apoptotic property and its role in the transcription of genes of both, host and pathogen, it appears to be an important component for targeting.
