عنوان انگلیسی I-19: In Vitro Derived Gametes: A Model for Studying Molecular and Cellular Biology of Germ Cells
چکیده انگلیسی مقاله Understanding the specification and development of mechanisms of germ cells is crucial to reproductive health including fertility, transmission of genetic diseases and cancer. Currently, it is estimated that one in seven couples are likely to experience difficulties conceiving. Several factors including gametogenesis dysfunction, congenital anomalies, endocrinopathies, immunological disorders and genetic diseases are known as causes of male infertility. Although recent advances in assisted reproductive technologies have given many infertile couples the opportunity to have their own child, there is potential for improvement in many of assisted reproductive procedures. The probability of success for both in vitro fertilization and donor insemination is relatively low and this decreases with age. This and other facts reflect our current knowledge and technical limitation for treating infertility. Generation of in vitro derived gametes is one of the techniques which hold a great deal of promise, as both scientific experimental models to understand molecular and cellular mechanisms underlying germ cell development and therapeutic approaches for study transmission of genetic diseases from one generation to the next and for treatment of infertility. We developed a new promoter based genetic selection of germline stem cells (GSCs). Germline stem cells, which can self-renew and generate gametes, are unique stem cells in that they are solely dedicated to transmit genetic information from generation to generation. In addition, we developed a strategy for the establishment of germline stem cell lines from embryonic stem cells. These cells are able to undergo meiosis, generate haploid male gametes in vitro and are functional, as shown by fertilization after intracytoplasmic injection into mouse oocytes. Molecular and cellular mechanisms underlying differentiation of ES to functional gametes should be elucidated in future research. In the proposed talk, we will discuss the current state of the filed, recent advances and future perspectives of this novel technology.
