چکیده انگلیسی مقاله Objective: MBD2 acts as a repressor on the human γ-globin gene. Also MBD2 has been shown to be associated with a complex known as MeCP1 which includes the NURD corepressor complex. Previous results from our lab suggest that MBD2 siRNAs can augment γ-globin expression in K562 cells. we shown before that MBD2 knockdown (35%) using siRNA in transient transfection result in Induction of γ-globin expression(1.7 fold).Although we have Enough level of infection(20%)by lentivirus system in K652 cells and this cell line expresses γ-globin gene,so it's a good choice for producing stably MBD2 knock down cell line. Materials and Methods: Five MBD2 shRNA-pLKO.1 vectors were screened in HEK 293T cells. Results: Two vectors gave ≥60% target transcript knockdown levels after normalization for transfection efficiency. These vectors were subsequently packaged and used to transduce K562 cells. Conclusion: We are in the process of determining the MBD2 and γ-globin expression levels of these cells using qRT-PCR.
