عنوان انگلیسی O-14: Genetic Aberrations in Phase I and Phase II Enzymes and Increased Risk of Heaand Neck Cancer
چکیده انگلیسی مقاله Head and neck cancer is the leading cause of cancer related deaths in Asian countries. Environmental as well as genetic factors are associated with cancer development. Carcinogens are detoxified by phase I and phase II enzymes like CYP1A1 and GSTM1. CYP1A1 is the phase I enzyme that detoxifies carcinogens in tobacco smoke whereas GSTM1 acts via conjugation with glutathione making carcinogen more electrophilic and easily excretable form the body. Genetic changes in these genes can potentially lead to the process of carcinogenesis. The current study was designed to investigate genetic changes of these genes in 437 head and neck cancer patients and 507 healthy controls using PCR-SSCP technique. The results demonstrated a novel frameshift mutation in CYP1A1 gene due to insertion of thymidine (T) at nucleotide 2842. Mean age of patients with CYP1A1 mutation was 51.75 (+15.7) years and seven percent patients had this mutation while no control showed this insertion of T. GSTM1 gene showed homozygous deletion polymorphism in significantly higher number of patients compared to controls (OD 4.3 and 95%CI of 2.9- 6.2). The mean age of patients with GSTM1 deletion was 49 (+11.9) years. These results suggest that the detoxifying genes may be altered leading to carcinogen activation resulting in head and neck cancer development. Mutations in CYP1A1 and GSTM1 gene may be one of the possible reasons for cancer progression and should be screened for gene therapy.
