P-77: Follicular Fluid Concentrations of Interleukin-6, Interleukin-8, TNF-α and Interleukin 10 in Polycystic Ovarian Syndrome Women

Background: While the etiology of PCOS has continued to remain a mystery, there can be found some evidence in support of the presence of chronic low-grade inflammation in women with this syndrome. One of the most significant manifestations of chronic inflammation is Pro-inflammatory cytokines. Therefore, the aim of this prospective study was to investigate the pattern of proinflammatory and anti-inflammatory cytokines in the follicular fluid (FF) of PCOS women undergoing in vitro fertilization (IVF). Materials and methods: Eighty women who met the selection criteria were recruited, and the FF was obtained during the follicular aspiration. Hence, we classify them as follows: (A) PCOS women; (B) Control women with normal ovulatory function who underwent IVF for treatment of tubal and/ or male infertility. Obtained FF from each patient was centrifuged at 300 g for 10 min. The supernatant was used to determine the concentration of a quantification of pro inflammatory cytokines interleukin (IL)-8 , IL-6, IL-10, and tumor necrosis factor-alpha (TNF-α) which was measured by the sandwich enzyme immunoassay ELISA based on a monoclonal antibody specific for them (Boster Biological Technology ltd, USA), according to the manufacturer's instructions. Results: After BMI was controlled, there was a significant increment in the concentration of inflammatory cytokines IL-8 (187 ± 43.01 pg/ml), IL-6 (29.39 ± 2.75 pg/ml) and TNF-α (6.35 ±1.7 pg/ml) in the FF of PCOS women undergoing controlled ovarian hyperstimulation versus control women(141.92 ± 28.87, 7.90 ± 0.28 and 4.02 ± 0.94 pg/ml respectively, P< 0.05). In addition, comparing PCOS and control group, we observed significantly lower levels of IL-10 concentration in PCOS women (4.02 ± 0.26 vs. 8.69 ± 1.10 pg/ml, P < 0.001). Conclusion: This result confirms the point that we can have the presence of a chronic low-grade inflammation and an involvement of the immune system in women with poly cystic ovarian syndrome. Therefore, it seems that targeting this inflammatory process by means of anti-inflammatory agents might be a good solution to the current treatment.
