P-217: The vascular Endothelial Growth factor VEGF+405 G/G Genotype May Influence Embryo Implantation in Assisted Reproductive Techniques

Background: Repeated implantation failure (RIF) is the main problem after using assisted reproductive techniques (ART). The main causes of RIF as a multifactorial problem including decrease in endometrial receptivity, defects of embryo or combinational. Successful embryo implantation depends on trophoblast proliferation, migration and invasion to the endometrium, all of which are associated with vascular endothelial growth factor (VEGF) as the major protein in stimulating of angiogenesis. This study aimed to determine the association betweenVEGF+405G/C polymorphism and RIF in infertile women. Materials and Methods: The patient group included 74 women with >3 RIF and the control group consisted of 149 healthy fertile women. Genotypes and allele frequencies of VEGF+405G/C polymorphism were determined by PCR-RFLP method and verified by Sanger sequencing Results: The frequencies of GG, GC and CC genotypes in patient group were 31.1, 48.6 and 20.3%, respectively while those frequencies in controls were 2.0, 47.0 and 51.0% respectively. The frequency of GG genotype was significantly higher in patients than that of controls (p< 0.001). CC genotype frequency was higher in controls than that of patients (p< 0.001). The frequency of GC genotype did not show any difference between groups. C as the wild allele was more frequent in controls while frequency of G as the mutant allele was higher in patients (p< 0.001). Conclusion: The VEGF+405 G/G genotype may influence embryo implantation and lead to RIF in ART candidates. Since this is the second report on association of this polymorphism with RIF, further studies in different ethnic population require determining this association.
