P-136: The Protective Effect of Vitamin C on Adverse Effect of Bisphenol A on The Ovary in Adult Rat

Background: Bisphenol A (BPA), an estrogenic environmental contaminant is also known for oxidative stressinducing effect. The aim of this study was to investigate the effect of vitamin C as an antioxidant on the ovary tissue in adult rats treated with BPA. Materials and Methods: Adult female rats (200 ±20 g) were divided into 4 groups (n=6):control, bisphenol A (60μg/Kg/day) , vitamin C (150 mg/Kg/day) and bisphenol A + Vitamin C. Oral treatments were performed for 20 days. After treatment, the right ovaries removed, fixed and tissue processing was carried out. Using stereological technique, the total volume of ovary, cortex, medulla, corpus luteum, the mean number of follicle, the volume of oocyte and its nuclei and thickness of zona pellucida were estimated. Data were analysed with SPSS using one-way ANOVA, and the mean difference was considered significant at the (p< 0.05). Results: Body and ovary weight showed no significant change in 4 groups (p>0.05). A significant decrease in the total volume of ovary, cortex, medulla and volume of corpous luteum in BPA group was found in comparison with the control group (p< 0.05). The mean number of antral follicle in group treated with BPA significantly decreased compared to control group (p< 0.05), while atretic follicle increased in this group. The volume of oocyte and its nuclei in antral follicles and also the thickness of zona pellucida in secondary and antral follicles indicated a significant decrease in BPA group compared to control group. Co-administration of vitamin C and BPA compensated for the adverse effects of BPA on the above parameters. Conclusion: This study showed that co-administration of vitamin C with BPA could prevent the adverse effects of BPA on the ovarian tissue in adult rat.
