Conditioned Medium Protects Dopaminergic Neurons in Parkinsonian Rats

Objective: Adipose stem cells (ASCs) secrete many neurotrophic factors and cytokines in the conditioned medium (CM) which protect neurons by its antioxidative and trophic effects. In this research, the expression of BDNF, NFG and NT4 genes and tyrosine hydroxylase positive (TH+) cells density and anti-oxidative activity of male Wistar rats lesioned by 6-hydroxydopamine (6-OHDA) and treated with ASCs-CM were assessed. Materials and methods: Experimental groups were: lesioned and sham rats, injected unilaterally by 20µg of 6-OHDA neurotoxin and PBS into the striatum respectively, another groups that injected intravenously with 3×106 cells (ASCs group), 500µl of CM (ASC-CM group) or medium (α-MEM group). All rats were evaluated with rotarod and apomorphine-induced rotation tests at 2, 4, 6 and 8 weeks post injection. At 8 week, some animals were sacrificed for real-time PCR , TH+ cells counting and oxidative stress activity. Results: It was a significant decrease of contralateral turns to the lesion of ASCs and ASC-CM groups as compared to neurotoxin lesioned or α-MEM groups at 8 weeks post transplantation. Cell and CM- injected rats showed a significant increase of staying on the rotarod as compared to lesion or α-MEM groups. Cell and CM-treated rats showed significant increase of NGF and NT3 genes, respectively compared with lesion. Both treated groups showed a significant increase of BDNF gene expression and TH+ cells density. Cell and CM transplantation induced an increase of GPX and SOD, with decrease of MDA activities in injured substantia nigra. Conclusion: Our current results suggest that ASCs and ASC-CM protect dopaminergic neurons through the expression of neurotrophin genes and anti-oxidative activity.
