Isolation, Characterization and Osteogenic Potential of Mouse Digit Tip Blastema Cells in Comparison with Bone Marrow- Derived Mesenchymal Stem Cells In Vitro

Objective: Limb regeneration which is mediated by blastema cells (BlCs) in mammals is limited to digit tip of neonates. Due to lack of access to BlCs in adults as well as the difficulty in isolating and expanding the BlCs from neonates, using a cellular population with similar features of BlCs would be a valuable strategy to direct non-regenerative wound towards regeneration. Therefore, this study first aimed to isolate and culture BlCs and explore their characteristics in vitro and secondly compare the capability of bone marrow-derived mesenchymal stem cells (BM-MSCs) as an alternative accessible cell source to BlCs for appendages regeneration. Materials and methods: BM-MSCs and BlCs were isolated from bone marrow and neonatal regenerating digit tip of C57B/6 mice, respectively. Then, they were characterized for the expression of cell surface markers using flow cytometry. Differentiation potential into skeletal cell lineages assessed by qRT-PCR and lineage-specific staining. Furthermore, the colony forming ability, proliferation, alkaline phosphatase activity, calcium content and osteogenic gene expression were evaluated in both BM-MSCs and BlCs cultures at day 7, 14 and 21. Results: qRT-PCR analysis revealed that the cells from both sources were readily differentiated into mesodermal lineages. The colony forming ability were significantly higher in BM-MSCs compared to those of BlCs (*P< 0.05). Alizarin red staining, calcium and ALP assay showed the same degree of mineral deposition in both BlCs and BM-MSCs. Gene expression level of osteblastic markers indicated the similar bone differentiation capacity for both BlCs and BM-MSCs in all time-points. Conclusion: It is believed that BlCs characteristics in vitro are similar to BM-MSCs therefore could be considered as BlCs substitute for regenerative approach with the potential of application in future clinical setting for regenerating human appendages.
