عنوان انگلیسی Melatonin Pretreated Blastocysts along with Calcitonin Administration Improved Implantation by Upregulation of Heparin Binding-Epidermal Growth Factor Expression in Murine Endometrium
چکیده انگلیسی مقاله Objective: Implantation failure is an obstacle in assisted reproduction techniques. A well-developed embryo and receptive endometrium are essential for successful implantation. Calcitonin is one of the molecules involved in uterine receptivity and embryo implantation. Moreover melatonin can promote embryo quality. So, to improve embryo quality and endometrial receptivity we examined the effect of pretreatment of blastocyst with melatonin along with administration of calcitonin on HB-EGF expression in pseudo-pregnant mice. Materials and methods: Two cell stage embryos were collected from the oviduct of 1.5 day pregnant NMRI mice and cultured to blastocyst stage in GTM medium contained 10-9M melatonin or nothing. Twenty four hours after calcitonin injection (2 IU), cultured blastocysts were transferred into the uteri of pseudo-pregnant mice. Two days after embryo transfer, implantation sites were counted and the level of HB-EGF mRNA and protein in uterine were evaluated by quantitative RT- PCR and western blot. Results: The results showed that melatonin pretreatment along with calcitonin administration significantly increased HB-EGF mRNA (p≤0.001) and protein (p≤0.001) in endometrium of pseudo-pregnant mice. Compared with control group (2.6 ± 0.5), the average number of implantation sites in the melatonin group (4.6± 0.5) and calcitonin group (7 ± 1) significantly increased (p≤0.05 and p≤0.001 respectively). There was a significant increase in implantation sites in melatonin and calcitonin combination group (8.6 ± 0.5, p≤0.001). Moreover calcitonin significantly enhanced calcitonin receptor mRNA (p≤0.001) and protein (p≤0.05) in uteri of both pregnant and pseudo-pregnant mice. Conclusion: In conclusion, melatonin pretreated blastocyst and calcitonin can increase HB-EGF expression in uterine of pseudo-pregnant mice
