چکیده انگلیسی مقاله Background: Given the global problem of antibiotic resistance among pathogens, researchers are looking for appropriate treatment alternatives to eliminate infections. The use of probiotics and their products can be a good alternative. Therefore, the inhibitory effect of cell-free supernatant (CFS) of probiotics against Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) was investigated.
Methods: The effect of CFS of eight strains of probiotic against E. coli and S. aureus was evaluated by well diffusion method. The agent that showed the highest inhibition diameter, it was selected to investigation of other antibacterial methods including MIC (Minimum Inhibitory Concentration), MBC (Minimum Bactericidal Concentration), and time-kill. SEM micrographs compared in treated and untreated bacterial cells with CFS of Lactobacillus acidophilus (SLA) LAFTI-L10 DSL. Finally, viability% of the Hu02 cells following 24,48 and 72 hours of incubation with various concentrations of SLA investigated.
Results: Among the tested strains, SLA showed the highest inhibitory diameter against E. coli and S. aureus (p ≤ 0.005). Also, the MIC of SLA was equal to E. coli and S. aureus (12.5 mg/ml) but different in MBC. In addition, almost 100% of bacterial cells removed after exposure to CFS (after 20 minute). Results of log CFU/mL demonstrated that SLA has bactericidal effect against S. aureus and E. coli. Toxicity assay showed that Viability% of the Hu02 cells was 31.71 to 81.09 after exposure 24, 48 and 72 hours.
Conclusion: SLA can be suggested as a suitable alternative to antibiotics due to its inhibitory effects on bacterial growth.
